Some stamp blocks (from Belgium) have a + sign at the top in the middle. (exactly as with a printer to calibrate the colors)
and at the bottom a number

Not with others.

For example, I have the block of Tintin and the moon in it twice my property, once without those characters and once with.

Does that make a difference in value or a difference for collectors?
